7 серийный анализ генной экспрессии
[лат. series — ряд; греч. analysis — разложение, расчленение; греч. genos — род, происхождение; лат. expressio — выражение]молекулярно-генетический метод, позволяющий одновременно количественно и качественно охарактеризовать экспрессию тысяч различных генов путем анализа их транскриптов. Два основных принципа лежат в основе С.а.г.э.: а) короткие нуклеотидные последовательности ДНК ("ярлыки") (10—14 п.н.), достаточные для идентификации индивидуальных генных продуктов, и б) конкатемеризация таких ярлыков (связывание их друг с другом в одну последовательность), что многократно увеличивает эффективность идентифицирования экспрессированной мРНК в библиотеках. Теоретически ярлык длиной 14 п.н., состоящий из составляющих любую нуклеотидную последовательность ДНК четырех видов нуклеотидных оснований, может определить 4 14 разных транскриптов, что многократно превосходит число генов, предположительно содержащихся в геноме человека и млекопитающих.
